You don't have to warm up the engine first. You'll still get AF into a cold empty block with the t-stat in place. Like you said though, if in doubt and it makes you feel better, pull a plug afterwards to check for AF.

When I had RWC engines, I drained, took temp sensor out and filled block, Then drained and filled again.. Thhe did same to manifolds. ALWAYS poke about with a coat hanger wire to make sure rust doesn’t block passages.
